Outline of Research at the Start

NOTCH受容体の活性化には、隣接した細胞間でリガンド分子が受容体分子と相互作用することが必要となり、活性化レベルに応じて、発現量は精密に調節される必要がある。これまでの研究より、NOTCH受容体を修飾している特殊なO型糖鎖修飾を変化すると、細胞膜での発現レベルが鋭敏に変化することを見出した。その一方で、糖鎖修飾が異なる異常なNOTCH受容体を小胞体で保持し分解する分子機構は不明である。本研究では、NOTCH受容体や構造的に類似のタンパク質を用いて、小胞体に存在する各種シャペロン分子との相互作用を中心に、分泌されずに分解に向かうNOTCH受容体の仕分け機構を明らかにし、その破綻により生じる病態を制御することを目指す。

Outline of Final Research Achievements

The present study provides a new perspective on glycobiology by demonstrating that O-glycans play important roles not only in protein folding but also in intracellular transport and secretion; the glycosylation map and functional analysis of DLK1 are also expected to have applications in understanding related diseases and drug discovery. Furthermore, it was suggested that O-glycan deficiency induces different transport and degradation pathways in DLK1 and NOTCH1, and it is expected that the sorting mechanism of O-glycan-mediated secretion and degradation will be elucidated in the future.

Academic Significance and Societal Importance of the Research Achievements

本研究は、O 型糖鎖がタンパク質フォールディングのみならず、細胞内輸送および分泌においても積極的に機能しているという新たな知見を提供し、糖鎖生物学の理解を一層深化させるものである。また、DLK1 の O 型糖鎖修飾マップの確立とそれに基づく機能解析により、DLK1 関連疾患の病態解明や糖鎖を標的とした創薬への応用が期待される。今後、同様の解析を Notch1 にも適用することで、O型糖鎖の分泌、もしくは、分解経路への仕分け機構の基本原理の理解と、O型糖鎖修飾が主役となるタンパク質の細胞内動態に関する新たなパラダイムが提示されることが期待される。
