Outline of Research at the Start

加齢黄斑変性や糖尿病網膜症などの治療成績は、抗VEGF療法により劇的に改善した。しかし、こうした病態の末期にみられる線維化に対しては未だ有効な治療法がない。我々が開発したペリサイト消失網膜症モデルマウスでは、糖尿病網膜症と同様の血管異常をきたした後に線維化をきたすことを見出している。筋線維芽細胞の周囲には多数のマクロファージが浸潤し、線維化を促進している様子がうかがえる。本研究では、網膜線維化を制御するマクロファージ亜集団を特定し、これを標的とした抗網膜線維化療法の開発をめざしている。

Outline of Annual Research Achievements

令和5年度の研究では、ペリサイト消失網膜の線維化を制御するマクロファージ亜集団を特定し、抗網膜線維化療法の標的となりうるかを検討した。生後1日マウスの腹腔内に抗PDGFRβ抗体を単回投与して、網膜血管壁のペリサイトを消失させた後、生後2週(急性炎症)および3週(慢性炎症)の網膜からCD45+CD11b+細胞分画をフローサイトメトリーにより精製し、シングルセルRNA-Seq解析を実施した。その結果、骨髄由来単球ではなく、活性化ミクログリアに由来するマクロファージ亜集団が線維化を促進していることが明らかとなった。

Strategy for Future Research Activity

次年度は米国National Institute of Health、National Cancer Institute、National Center for Advancing Translational Sciencesの研究チームが合成した低分子化合物を用いて、線維化促進マクロファージを標的とする抗網膜線維化療法の可能性を検証する。
