Outline of Research at the Start

免疫系による炎症反応は生体防御に必須であり、加齢によって炎症反応がうまく調節できなくなると、生体防御機能の低下と病的炎症のリスク増大に繋がる。近年DNAメチル化異常と病的炎症の関連が注目されているが、発症機構は未だ不明のままである。DNAメチル化異常は老化によって蓄積され、TET遺伝子の変異によって異常がさらに加速することが知られている。
本研究では、TETを欠損させた実験マウスモデルを用いて、人工的に急激なDNAメチル化異常を誘導し病的炎症が起こる機構を解明することを目的とする。これにより病的炎症発生機構の一端が解明され、炎症性疾患の新規診断法や治療法の開発に繋がることが期待される。

Outline of Final Research Achievements

In this study, we aimed to address the academic question: “Why does aberrant DNA methylation lead to immune dysregulation and pathological inflammation?” Using a replicative senescence model, we found that inflammatory macrophages exhibited a loss of heterochromatin, which is normally localized near the nuclear envelope. In addition, the inflammatory macrophages induced in our model mice exhibited characteristics similar to those of macrophages in human ECRS (eosinophilic chronic rhinosinusitis) patient samples. In a T cell-specific TET2/3-deficient model, we observed an increase in cell populations such as NKT cells, CD4+ cytotoxic T lymphocytes, and T follicular helper cells. Interestingly, the increased NKT cells exhibited strong anti-tumor activity. Thus, we are currently exploring their potential application in cancer immunotherapy. Additionally, we developed a novel algorithm, the GhmCNs method, and demonstrated its utility in the identification of previously unknown enhancers.

Academic Significance and Societal Importance of the Research Achievements

本研究のモデルマウスで誘導した炎症性マクロファージと、ヒトECRS患者サンプル中のマクロファージは似た性質を持つことを見出し、今後の新規治療法の開発に繋がりうる社会的意義のある研究成果を出すことができた。予想外の成果として、炎症性のT細胞中に抗腫瘍活性の強いものを見出し、現在がん免疫療法への応用を模索している。また、独自のアルゴリズムGhmCNs法を開発し、既知のエンハンサー領域での確認、未知のエンハンサー探索への応用などの可能性を提示できたことは、学術的に意義のある研究成果と言える。
