Outline of Research at the Start

パーキンソン病(PD)は病初期からalpha-synuclein(aSyn)が自律神経や嗅神経に蓄積し、脳幹・辺縁系・大脳皮質に伝播する全身疾患で、進行を抑制する疾患修飾療法はない。aSynが蛋白分解系オートファジーで分解されることから、本研究ではオートファジー活性を「神経細胞内で集約・局在しaSyn分解を促進する」という治療薬開発に挑む。オートファジーを活性化するアクロレインとその誘導体ライブラリーを作製している。本化合物群等を用いて高速自動画像スクリーニングからヒット群を同定する。そのヒット群の中から自律神経細胞、黒質ドパミン神経細胞、マウス大脳の全モデルで薬効を示す創薬シーズを同定する。

Outline of Final Research Achievements

Parkinson’s disease is a systemic disorder in which alpha-synuclein (aSyn) accumulates in the autonomic and olfactory nervous systems from the early stages, subsequently propagating to the brainstem, limbic system, and cerebral cortex. Currently, no disease-modifying therapy exists to slow its progression. Since aSyn is degraded via the autophagy-lysosomal pathway, this study aimed to develop therapeutic agents that “promote aSyn degradation by inducing autophagy activation and its aggregation/localization within neurons.” In 2022, our group identified acrolein, an endogenous compound that induces autophagy (EMBO J, 2022). Subsequently, we screened a library of 1,200 approved drugs and identified six novel compounds with similar mechanisms of action (eLife, 2024).

Academic Significance and Societal Importance of the Research Achievements

研究期間全体で明らかにした成果・社会的意義は下記の点に集約される。
・酸化ストレス時にのみ誘導されるオートファジーにおいて特異的に働くリソソーム分布調節機構を特定したこと。
・上記の特異的機構によるオートファジーを誘導する新規既存薬を6種同定し、その中の3種は血液脳関門を通過することから、パーキンソン病治療薬としてのポテンシャルが高く、根本的治療法開発に繋がる可能性を秘めること。
