Outline of Research at the Start

申請者は、チェレンコフ光閾値以下のエネルギーの放射線照射により水が発光する現象を発見した。この紫外線は発光には紫外線が多く含まれ、細胞中のDNA損傷を引き起こしていると予想している。この点を明らかにするために、260nm付近の紫外線領域まで測定可能な分光画像測定装置を開発することで、放射線照射による水の発光がDNA損傷を引き起こしていることを明らかにする。また、この発光に含まれる紫外線でラジカルを発生させる放射線増感剤、および紫外線の影響を減らす放射線防護材の開発を行い、放射線照射による水の発光現象が生体に影響を与えていることを間接的に証明し、また発見した水の発光現象の医療応用を進める。

Outline of Annual Research Achievements

チェレンコフ光閾値以下のエネルギーの放射線照射による水の発光がDNA損傷の原因であることを明らかにするために、紫外線が水発光に多量に含まれることを実測により明らかにした。
そのために、まず高い波長分解能を有する超高感度分光画像測定装置を開発した。スペクトル計測はバンドパスフィルターを暗箱の外から蓋を開けることなく交換可能とし、効率的な高波長分解能分光画像測定を実現した。
この開発した装置を用いることで、水の発光スペクトルを高分解能で得ることに成功した。その結果、紫外線が水発光に多量に含まれることを明らかにし、また300nm付近の発光を多く含むことを明らかにした。この発光を利用すれば、放射線の増感効果を実現できる可能性のあることを確認できた。
また、水の発光現象の発光機序と性質を理論計算とモンテカルロシミュレーションを用いて明らかにすることを試みた。
発見した放射線照射により発生する水の発光に含まれる紫外線を用い、酸化チタン(TiO2)ナノ粒子との反応で生じるラジカルにより腫瘍細胞を殺傷することを試みた。申請者の発見した光は、チェレンコフ光と違い、コヒーレントな光ではないため、発生後に位相の異なる光同士が打ち消しあい、光の強度が著しく減少する。しかし、発光点の近傍にTiO2ナノ粒子が存在する場合、打ち消しあう前にTiO2ナノ粒子と反応し、ラジカルを多量に生成する可能性がある。本実験は、現在継続中である。

Reason

高分解能発光波長計測装置の開発は終了し、水の発光中に多くの紫外線が含まれていることを実証できた。また、関連研究において多くの研究成果がでており、査読付き論文が発表出来ている。

Strategy for Future Research Activity

(1)今年度は、酸化チタンの実験に加えて、5-ALAと放射線照射発光を用いた放射線増感効果の細胞実験での実証を行う。申請者の発見した水の発光に含まれる紫外線と5-ALAの反応により生じるラジカルが、腫瘍の治療効果を著しく高めることを、培養腫瘍細胞(脳腫瘍細胞C6 glioma、膵臓がん細胞PANC-1)を用いて、実証する。
(2)またビタミンB2と放射線照射発光を用いた放射線防護効果の細胞実験での実証を行う。申請者の発見した水の発光に含まれる紫外線がビタミンB2により、長波長に変換されることを利用し、放射線影響を低下させることを、培養正常細胞を用いて評価し、実証する。
