| Outline of Research at the Start |
本研究は、メチル化(5mC)の異常に起因するインプリンティング異常症において、ヒドロキシメチル化(5hmC)が病態にどのように関与しているかを解明することが目的である。5mCと5hmCを区別するための酸化バイサルファイト処理に続いてゲノムワイドプロファイリングを行い、“hydroxymethylome”を明らかにする。さらに脳サンプルの解析や、疾患特異的iPS細胞を用いたin vitroモデルに対して5mC→5hmC変換酵素TET1-3のゲノム編集ノックアウトと神経分化誘導を行い、インプリンティング異常症における神経症状の病態解明と、化合物スクリーニングによる治療薬の発見を目指す。

| Outline of Final Research Achievements |
This study aims to elucidate the role of hydroxymethylation (5hmC) in the pathogenesis of imprinting disorders caused by abnormalities in DNA methylation (5mC). To date, we have completed oxidative bisulfite treatment, pyrosequencing, and methylation bead array analyses. During the course of analysis, we identified several novel imprinting disorders, including ZNF597 methylation disorder and maternal uniparental disomy of chromosome 6. We are also developing a method for analyzing 5mC and 5hmC using nanopore sequencing without bisulfite treatment. In the future, we aim to further advance this method to elucidate the molecular mechanisms underlying the onset of neurological symptoms in imprinting disorders and to identify target molecules involved in disease pathogenesis.

| Academic Significance and Societal Importance of the Research Achievements |
不可逆的なゲノム配列とは対照的に、その修飾機構であるエピゲノムは可塑性を持ち、治療の標的となる可能性がある。本研究により、ヒドロキシメチル化がインプリンティング異常症の発症に関与している可能性が示唆された。今後、その詳細なメカニズムを解明し、その可塑性を活かしたエピゲノム治療薬の開発を目指す。さらに本研究の成果は、先天異常症候群に加えて、生活習慣病や悪性腫瘍、精神疾患などの後天的なメチル化異常、エピゲノム異常に起因する疾患についても診断法・治療法の開発の手がかりを与え、再生、生殖、精神医学といった多方面の生命科学分野に波及し、国民健康の増進に広く貢献することも期待される。
