骨の恒常性は破骨細胞による骨吸収と骨芽細胞による骨形成のバランスで維持される。破骨細胞分化に必須のサイトカインであるRANKLは、肥大軟骨層軟骨細胞、骨芽細胞系細胞、脂肪細胞前駆細胞、骨細胞などが発現し、ライフステージさらには病的状況下によっても多様に変化することが知られている。本課題では、間葉系支持細胞におけるRANKLの空間的発現解析技術を駆使することで、骨リモデリングの起点となり得るRANKL発現細胞の時空間的分子制御機構と、RANKL発現間葉系細胞の細胞性質と運命経路の解明するとともに、最終的に病的RANKL発現機構に特異的な制御システムの分子基盤を明らかにする。

| Outline of Final Research Achievements |
Bone homeostasis is maintained through the balance between osteoclast-mediated bone resorption and osteoblast-mediated bone formation. RANKL is an essential cytokine for osteoclast differentiation, expressed not only in a membrane-bound form but also cleaved extracellularly to produce a soluble form. Through analyses of mice lacking soluble RANKL, we have demonstrated that direct cell-to-cell contact between osteoclast precursors and mesenchymal cells producing membrane-bound RANKL is necessary for osteoclast differentiation in vivo. In this research project, we focused on mesenchymal cells as a source of RANKL, which changes with life stage and pathological conditions, and investigated their cellular properties and the spatiotemporal mechanisms regulating RANKL expression.

| Academic Significance and Societal Importance of the Research Achievements |
高齢化社会では、骨の恒常性破綻がQOL低下や要介護状態に直結する重要な課題である。間葉系細胞におけるRANKL発現制御機構の解明により、様々な骨量減少疾患に応じた新たな治療標的の創出が期待される。また現在、RANKL阻害薬であるデノスマブは高い骨吸収抑制効果を示す一方で、休薬後の急激な骨密度低下(オーバーシュート現象)が問題視されている。加齢や疾患に伴う骨量減少の分子機構、特にRANKL発現細胞動態に関する理解を深めることで、デノスマブが抱える課題を克服し、より安全で効果的な個別化治療戦略の開発にも貢献できる。以上より本研究成果は国民の健康増進、QOL向上、医療費削減に繋がると考えられる。
