Outline of Research at the Start

加齢,性差,循環特性,運動に対する血圧応答(運動昇圧応答)など脳血管シェアレイト応答に影響する生理要因を同定することにより,運動効果の個人差やばらつきの生理メカニズムを解明することを本研究の目的とする.脳血管シェアレイトに影響を及ぼす生理要因を調査し,特に脳血管内皮機能に影響する加齢,性差,血管特性(脳血管コンプライアンス等),運動昇圧応答の差異が脳血管シェアレイト応答に及ぼす影響を確かめる.これらの要因が脳血管シェアレイトに影響するとすれば,個人の生理的特性の差異により最適な運動が異なることが示唆される.

Outline of Annual Research Achievements

運動による脳循環機能及び認知機能改善効果は疫学調査等から疑いの余地はない.しかしながら,運動効果の個人差や運動のこれら改善効果の生理メカニズムは未だ十分に明らかにされているとは言えず,適切な運動処方構築のハードルとなっている.本研究課題では,申請者が行ってきた関連研究の知見から,脳循環における運動効果のキーファクターとして運動に対する脳血管シェアレイト応答に焦点をあてた.運動様式の違いによる脳血管シェアレイトの応答の差異が脳血管内皮機能,さらに認知機能に直接的に影響するのか?また加齢,性差,血管特性,運動昇圧応答等の脳循環動態に及ぼす生理要因の個人差が脳血管シェアレイトにどの様な影響を及ぼすのか?という2つの学術的問いに関連して,2つの研究課題に取り組む.本年度は2年目であり研究課題1として,運動に対する脳血管シェアレイトの応答と運動効果(脳血管内皮機能,認知機能)との関連性を明らかし,脳血管シェアレイトの応答を運動による脳循環,脳機能の改善の指標と捉えられるか検証した.幾つかの関連論文の発表に加え,運動中の運動昇圧応答及び脳血管シェレイトの差異による脳血管内皮機能に関する実験を行った.運動中の血管シェアレイトの差異による脳血管内皮機能へ及ぼす影響を調査した実験はニュージーランドのオークランド大学Fisher教授との共同研究として行った.これらの実験結果は,学会等で発表し,また1編は原著論文としてまとめ,脳循環関連雑誌に投稿中である.シェアレイトの差異に関する国際共同研究の実験については,データ分析が終了したところであり,現在論文と投稿準備中である.

Current Status of Research Progress

1: Research has progressed more than it was originally planned.

Reason

関連研究は多数行っており発表論文は本年度16編であった.特に研究課題に関する主要研究として2つの実験を行った.我々の先行研究では,運動の様式により脳への血管シェアレイト応答が大きく異なること,特にインターバル運動では脳の血管へのシェアレイトが高くなることを報告していた.この結果に興味を持った,ニュージーランドのオークランド大学Fisher教授との共同研究として,この運動様式による脳血管シェアレイトの違いが脳血管内皮機能に影響するかを調査した.脳血管シェアレイトが高いインターバル運動で脳内皮機能が改善するとの仮説を立てたが,結果は異なっていた.現在,データの分析は全て終わっていないため終了次第,論文投稿準備を行う予定.また,他の研究では、冷水刺激による昇圧応答によるシェアレイト増加が,血管応答に及ぼす影響を検討した.冷水刺激時の頸動脈血管応答が心臓循環器疾患リスクになることが示唆されているが,内皮機能に関連すると示唆されておりそのメカニズムを調査した.実験は終了したが,データを分析中である.一方、研究課題2である,昇圧応答の個人差が及ぼす影響を調査する研究は,前倒しして行っており重要な知見が得られている.例えば,運動昇圧応答には個人差があり,昇圧応答の違いと運動による認知改善効果を調べたところ,過度な運動昇圧応答を有する個人は,運動による認知機能改善効果が消失することが明らかとなった.この結果は,学会で発表し,現在原著論文にまとめて脳研究の雑誌に投稿中である.以上,研究計画は,前倒しして研究結果も得られている状況である.

Strategy for Future Research Activity

本研究課題では,申請者が行ってきた関連研究の知見から,脳循環における運動効果のキーファクターとして運動に対する脳血管シェアレイト応答に焦点をあて,運動に対する脳血管シェアレイトの応答と運動効果(脳血管内皮機能,認知機能)との関連性を明らかし,脳血管シェアレイトの応答を運動による脳循環,脳機能の改善の指標と捉えられるか検証(研究課題1),また,脳血管シェアレイトの増加に及ぼす要因(加齢,性差,血管特性,運動昇圧応答等)との関連性から,運動効果の個人差の生理メカニズム(脳血管シェアレイトの応答の差異)の解明(研究課題2)を本研究の目的として実験を行ってきた.現在は,研究機関の半分が終了したところであるが,既に研究課題2の運動昇圧応答の個人差に関する実験を行い論文投稿にまで進んでいる。今後は、研究課題2について,加齢や性差による血管特性や運動昇圧応答の差異による運動中の血管シェアレイトの違いがどのように運動効果(血管内皮機能や認知機能など)に影響するか調査する.また関連研究を進めていく.
