Outline of Research at the Start

1)一軸配向化コラーゲン面上での2次元培養が、2種の線維芽細胞のECM関連遺伝子発現に対して与える効果、2種のMSCの腱性線維芽細胞への分化とECM関連遺伝子発現に対して与える効果を、無配向コラーゲン面と比較して明らかにする。
2)一軸配向化コラーゲン線維を3次元化した線維束およびそれにGAGを複合化した線維束を用いた3次元培養が、MSCの腱性線維芽細胞への分化とECM関連遺伝子発現に対して与える効果を2次元培養のそれらと比較して明らかにし、またそこに介在するGAGの役割を示す。
3)一軸配向化コラーゲン線維束のin vivo移植実験を行い、vitroとvivoにおける結果の相同性を確認する。

Outline of Final Research Achievements

In this study, we performed following experiments using collagen microfibers comprising aligned fibrils. We developed a unique spinning technique for collagen microfibers in which the material is stretched in critical fibrillogenesis concentration solutions. This process resembles melt spinning, in which the plasticized resin is stretched and solidified, resulting in a narrow and durable fiber with oriented molecules. The spinning process demonstrated exceptional productivity and the ability to generate collagen microfibers with diameters of approximately 20 μm, featuring uniaxially aligned collagen fibrils not only on the surface but also within the core of the microfibers. The excellent mechanical properties of the dry microfibers reflected those of the microfiber bundles under wet conditions. We clarified the effect for differentiation of bone marrow stem cells and mechanism of extracellular matrix production inducted by collagen microfibers comprising aligned fibrils.

Academic Significance and Societal Importance of the Research Achievements

先行研究に基づき独自に着想した一軸配向化合成コラーゲンの構成と組織構造がMSCの分化を誘導するという仮説の検証は、一軸配向化コラーゲンに関するCell-material interactionを解明する研究であり、高い学術性がある。本研究の成果は、生体において移植自家腱が誘導する腱・靱帯再生・再構築現象の分子機序に関する核心部分の解明に貢献すると共に、高靱性一軸配向化合成コラーゲン線維束を使用した「生体内でリモデリングを受けて再強化される人工腱」を世界に先駆けて開発する道を開く。
