Outline of Research at the Start

タンパク質は、環境(pH、温度)に応答して活性が変化する。タンパク質の活性温度は結晶化温度よりも高い場合がほとんどであるが、従来のX線結晶構造解析においては、20℃以下の制限された条件でのみ、結晶の質を保持したまま時分割実験などを実現していた。活性温度よりも低い温度で得られた構造情報のみで、果たしてタンパク質の機能を解明する情報は十分なのだろうか？
本研究では、10～45℃程度の温度環境において、リアルタイムで結晶内のpHを変更しながら回折実験を行い、変化する構造情報を解析し、温度が影響する活性状態の構造と反応機構の相関を明らかにすることを目的とする。
