Outline of Research at the Start

アルツハイマー病発症機構において何故アミロイド蓄積がタウ病理をひきおこすのか不明である。本研究では、Aβ蓄積依存的にタウ病理を再現するマウスモデルを用いてそのメカニズムを解明することが目的である。これによって、アルツハイマー病を予防・治療するための新たな標的が見いだされる。本研究の最終目的は「アルツハイマー病のない世界を創る」ことである。

Outline of Annual Research Achievements

これまで、Aβアミロイドーシスがタウ病理を促進するというモデルは存在したが、我々は「Aβ蓄積がなければタウ病理が見られない」という画期的モデルの作製に成功した。このようなモデルの開発は世界で初めての成果である。現在、共同研究者らが本モデルの微視的トランスクリプトーム解析(spatial transcriptomeおよびin situ RNA sequencing)を行っており、Aβ蓄積依存的にタウ病理を惹起する因子の解析が進んでいる。

現在まで使用してきたモデルよりもさらに早期にタウ病理を示すモデルの作製に成功したので、研究が加速されつつある。

Strategy for Future Research Activity

微視的トランスクリプトーム解析によって「Aβ蓄積依存的にタウ病理を引き起こす因子」を絞り込む。これらの因子に対してウィルスベクターを用いてノックダウン実験を行い、因果関係を樹立する。
