Outline of Research at the Start

最近、難治性がんにおいて変異KRASシグナルによって鉄中毒が引き起こされることが明らかになった。そこで、本研究では、鉄代謝異常による鉄中毒状態を利用して、代表的ながん遺伝子のRAS変異がんの代謝特性である鉄中毒を利用した創薬研究を目指す。すなわち、①RAS変異がんにおける鉄代謝異常の検証：自由鉄、ヘム鉄の役割の解明、②鉄中毒を標的とする創薬：二価鉄または遊離ヘム鉄で活性化する独自のプロドラッグの開発、③フェロトーシス誘導による治療増感、二価鉄またはヘム鉄蛍光センサーによる鉄代謝調節物質のスクリーニング及びドラッグリポジショニングの検討を行う。
