Outline of Research at the Start

独自に開発したアリールケトン型3CLプロテアーゼ阻害剤 YH-53 から新規不可逆的阻害剤を誘導し、これを基に新型コロナウイルス感染症（COVID-19）治療薬の創製を目指す研究である。具体的には、3CLプロテアーゼを駆逐できる新規阻害分子の創製を実施する。そのため、先ず可逆的阻害剤YH-53から不可逆的阻害剤への変換を行う。3CLプロテアーゼの活性中心に特異的に反応し、当該酵素を完全に失活させるが、一方で生体分子との反応性を極力抑えることができる安定な構造を見出す。さらに得られた阻害剤をPROTACなど複数のデグレーダーと架橋することで、ハイブリッド型阻害剤に導く。
