Outline of Research at the Start

研究代表者はこれまでに慢性アレルギー性気道炎症を制御する分子機構に関して、分子・細胞・生体レベルでの研究を一貫して行ってきた。本研究では、難治性アレルギー炎症の病態形成の上流に位置する病原性Th2(pathogenic Th2; Tpath2)細胞が生体内で分化を誘導される分子機構を明らかにすることを目的とする。特に「生体内でTh2細胞がTpath2細胞としての病原性を獲得し、長期間にわたってその病原性が維持される分子機構」を細胞レベルおよび生体レベルで解明する。

Outline of Annual Research Achievements

研究代表者はこれまでに慢性アレルギー性気道炎症を制御する分子機構に関して、分子・細胞・生体レベルでの研究を一貫して行ってきた。本研究では、難治性アレルギー炎症の病態形成の上流に位置する病原性Th2(pathogenic Th2; Tpath2)細胞が生体内で分化を誘導される分子機構を明らかにすることを目的としている。特に「生体内でTh2細胞がTpath2細胞としての病原性を獲得し、長期間にわたってその病原性が維持される分子機構」を細胞レベルおよび生体レベルで解明を目指している。具体的には、1. 細胞内でのオートファジーによる脂肪滴分解機構であるリポファジーに着目し、Tpath2細胞(Immunity 2018, Nat Immunol. 2019, Immunity 2022)の病原性機能獲得を担う分子機構を解明、2. 慢性炎症時に肺の組織中に誘導される異所性リンパ組織の一種であるinducible bronchus-associated lymphoid tissue(iBALT)を構成する線維芽細胞の亜集団が、Tpath2細胞の病原性を維持する機構について、線維芽細胞の1細胞レベルの網羅的解析を通じて制御因子の同定を目指す。3. ヒト(患者)の組織及び細胞で、最先端解析技術を用いて検証(Proof of Concept:POC)を行う。
我々は、これまでの検討でマイクロ・リポファジーを介したTpath2細胞誘導の分子機構の一端を解明し、現在論文をRevise中である。

1: Research has progressed more than it was originally planned.

Reason

我々は、これまでの検討で(1)マウスでHouse Dust Mite(HDM)の頻回曝露で引き起こした炎症肺組織中において、異所性リンパ組織が誘導され、同部位で複数の長鎖不飽和脂肪酸の濃度が上昇すること、(2)マウス炎症肺中のTpath2前駆細胞で、脂質代謝に関与する複数の遺伝子群が高発現すること、(3)Tpath2前駆細胞で、細胞質に多数の脂肪滴とリソソームの融合(=マイクロ・リポファジー)が認められること、をマウスおよびヒト検体(好酸球性副鼻腔炎患者検体)の解析から見出した。さらに、マイクロ・リポファジー関連分子について、阻害剤を用いて阻害することやCD4 T細胞特異的な遺伝子欠損マウスでTh2細胞のST2発現が抑制されることを明らかにした。マイクロ・リポファジー依存的なST2発現上昇は、脂質代謝を司る転写因子がマイクロ・リポファジーによって活性化されST2遺伝子座に直接結合することによる、という一連の分子機構も明らかにした。また、ヒトにおいて、同様の機構が存在することをヒトECRS患者の鼻ポリープ組織及び末梢血検体を用いて解析した。以上、当初計画した以上に順調に研究が進んでいる。さらに、上記の研究とは他に、好酸球性食道炎の組織線維化について病態形成機構を解析し、Amphiregulinを産生する病原性ヘルパーT細胞がその病態形成機構に関与することを明らかにした(Kaneko et al. JACI Glob. 2024)。

Strategy for Future Research Activity

1. “Tpath2細胞”におけるリポファジーを介した病原性獲得機構の解明
今回、我々がこれまでに同定した、Tpath2細胞(Nakayama et al. Annu Rev Immunol 2016, Morimoto Y et al. Immunity 2018, Okano M et al. Immunity 2022)について、同細胞集団のin vivoにおける分化および誘導に必須の機能分子(群)を探索した。特に、Tpath2細胞がIL-33刺激を受け取るために必須であるIL-33受容体分子のST2を生体内で高発現させる細胞・分子機構の解明を目指した。その結果、我々は、慢性アレルギー性気道炎症を引き起こしたマウス肺では、(1)長鎖不飽和脂肪酸が増加すること、(2)炎症肺のTh2細胞が、長鎖不飽和脂肪酸を取り込んで、脂肪滴が細胞質に形成されること、(3)この脂肪滴からリポファジー、リポライシス経路を介して脂肪酸代謝が活性化すること、(4)その結果下流の転写因子を介してST2の発現が誘導されること、を見出した。さらに、我々は、ヒトにおいても同様の機構が存在し、慢性アレルギー性炎症の病態形成に関与することを見出した。今後は、どのような分子・細胞機序を介して、炎症局所での長鎖不飽和脂肪酸増加が起こるのか、その詳細を解析していく。
