Outline of Research at the Start

卵巣明細胞癌は化学療法抵抗性で新規治療法の開発が期待されている。本研究では、卵巣明細胞癌のマウスモデルを用いて、卵巣明細胞癌で高発現しているサイトカインであるIL-6が腫瘍免疫に与える影響を調べて、IL-6の抑制が免疫療法の効果を増強させるかを検討する。また、これまでの解析によって、卵巣明細胞癌ではIL-17発現が腫瘍免疫活性と強く相関することが明らかになった。本研究では、IL-17の投与によって免疫療法の効果が増強するかを調べる。

Outline of Annual Research Achievements

ヒト卵巣明細胞がんにおいて、IL-17の高発現が腫瘍免疫活性化と相関することを見出した。そして、マウス卵巣明細胞がんモデルを用いて、IL-17投与が腫瘍免疫活性化をもたらすこと、そしてIL-17を産生するRORg(+)CD4+T細胞の移入が抗PD-1抗体による治療効果を増強させることを見出した。また、IL-6の発現抑制は、免疫不全マウスでは軽度に腫瘍増殖を抑制するが、免疫能を有するマウスでは顕著に腫瘍増殖を妨げることを見出した。それらの研究については、現在実験を続けている。
卵巣癌や腫瘍免疫に関連する論文としては、(i) 卵巣明細胞がんにおいて、YAP1の低下がフェロトーシス抵抗性と予後悪化に関わっていること(Furutake Y, Mol Cancer Ther 2024)、(ii) 卵巣高異型度類内膜がんは病理組織形態的に卵巣高異型度漿液性がんと類似しているが、分子的には全く異なる腫瘍であること(Takamatsu S, Br J Cancer 2024)、(iii) 卵巣高異型度漿液性がんの病理サブタイプ分類のためのAIアルゴリズム(Ueda A, Am J Pathol 2024)、(iv) 子宮内膜がんにおいてtertiary lymphoid structureが予後良好や、免疫チェックポイント阻害剤の効果と関連していること(Suzuki H, Cancer Immunol Immunother 2025)を報告した。さらに、子宮内膜がんの腫瘍免疫(Murakami K, Expert Opin Investig Drugs 2025)や、卵巣がんのDNA相同組み換え修復と薬剤感受性の関連(Takaya H, Expert Rev Mol Diagn 2025)に関する総説論文を出版した。

Strategy for Future Research Activity

(i) マウス卵巣明細胞がんモデルにおいて、IL-17がNF-kBを介して、腫瘍細胞からのサイトカイン分泌を促すかを明らかにする。
(ii) マウス卵巣明細胞がんモデルにおいて、IL-6が腫瘍免疫に影響を与えることで、腫瘍の増大を促すかを明らかにする。
