Outline of Research at the Start

腸内細菌叢が運動に対する炎症・ストレス応答やコンディション変化の個人差を規定するという仮説のもと、腸内細菌叢を用いたコンディショニングの個別化に向けた研究を行う。まず、一過性運動に対する炎症・ストレス応答の個人差を調べ、その個人差を規定する腸内細菌を明らかにする。次に介入研究を行い、プレ・プロバイオティクスによるコンディション改善効果を規定する腸内細菌を明らかにする。これらの研究で得られた知見に基づき、腸内細菌叢を用いたストレス応答の予測モデルやプレ・プロバイオティクスによるコンディション改善効果の予測モデルの構築を目指す。

Outline of Annual Research Achievements

本研究課題では、腸内細菌叢が運動に対する炎症・ストレス応答やコンディション変化の個人差を規定するという仮説のもと、腸内細菌叢を用いたコンディショニングの個別化に向けた研究を行う。2023年度は、腸内細菌叢解析プロトコルを確立した上で、一過性運動に対する炎症・ストレス応答の個人差に関連する腸内細菌叢の特徴を明らかにするための研究に着手することを目標として研究を行った。

先行研究で報告された腸内細菌叢解析プロトコルを精査し、以下の通り糞便採取から群集構造解析までのプロトコルを決定した。1) 糞便採取:グアニジンチオシアン酸塩溶液入り採便容器を用いて糞便を採取し、DNA抽出まで常温で保存する。2) DNA抽出:糞便採取後1週間以内に、ビーズ破砕とスピンカラムを用いてDNA抽出を行う。3) ライブラリー作成:16S rRNA遺伝子のV4領域をアダプター付きプライマーを用いてPCR増幅後、磁性ビーズによりPCR産物を精製する。インデックス付きプライマーを用いたPCRにより、精製されたPCR産物にインデックスを付加し、再度磁性ビーズにより精製する。4)DNAシーケンス:次世代シーケンサーiSeq100を用いてDNAシーケンスを行う(24~48サンプルをマルチプレックスする)。5) リードのクオリティーコントロールとトリミング:FastQCとBBDukを用いて、リードのクオリティーコントロールとトリミングをそれぞれ行う。6) 群集構造解析:菌叢解析パイプラインQIIME2を用いて行う。
実際に上記のプロトコルを用いて解析を行ったところ、良好な結果が得られたため、今後はこのプロトコルを用いて腸内細菌叢の解析を行うことにした。

また、一過性運動に対する炎症・ストレス応答の個人差に関連する腸内細菌叢の特徴を明らかにするための研究について倫理申請を行った。

本研究で用いる腸内細菌叢解析プロトコルを確立し、一過性運動に対する炎症・ストレス応答の個人差に関連する腸内細菌叢の特徴を明らかにするための研究について倫理申請を行ったが、実験開始には至らなかったため。

Strategy for Future Research Activity

2024年度は、一過性運動に対する炎症・ストレス応答の個人差に関連する腸内細菌叢の特徴を明らかにするため、年間を通じて実験を行う。また、高強度・高用量のトレーニングによるコンディション悪化の個人差に関連する腸内細菌叢の特徴を明らかにするために、本学の体育各部に研究協力を打診し、研究の準備を進める。
