Outline of Research at the Start

澱粉資源植物サゴヤシは,不良環境に適応し,食用澱粉の生産,アレルギー対応食品の製造,バイオ燃料の原料として期待される一方,withコロナのニューノーマル社会やSDGsで需要が高まっている。本研究は,主に自然林から収穫していた本種に,資源維持のコンセプトを導入し,積極的な新植で増産を図り,持続的プランテーション農業技術の実践に貢献する。具体的には [1] 難発芽種子の発芽勢改善による実生育成,[2] 実生苗への窒素固定菌,アーバスキュラー菌根菌の接種による生育促進技術開発,[3] 吸枝移植では,2年後には40%以上が枯死する実情を改善するため,バイオコントロール技術の導入による生存率向上を図る。

Outline of Annual Research Achievements

本研究では,次の3項目のテーマについて小項目を設定し,5年間で順次調査解析を進める計画である。
2024年度は上半期と下半期に1度ずつ,述べ4名の現地訪問による調査・試験を実施するとともに,国際研究集会で3度の基調講演,1度の招待講演,6件の発表,国内研究集会での2件の発表,2編の論文公表による成果発表を行なった。
(1) 難発芽種子の発芽勢改善等による移植苗育成技術の確立:インドネシア・パプア州で得た有刺タイプ民俗変種の果実より,発芽抑制物質を含む果皮や肉質種皮を外した後に,種子の胚上部に位置する朔蓋を除去することで発芽日数を短縮できることを確認できた。また,種子から発芽した実生個体より得た部位を用いた組織培養に取り組み,培地へのココヤシ果実の液状胚乳と炭の添加が新組織の分化と発育に有効であるとの結果を得た。
(2) 移植苗への有用微生物等の処理による生育促進:インドネシア・リアウ諸島州テビンティンギー島の泥炭層の深さが異なる地区(深い泥炭:5m>,中間:4 - 5m,浅い泥炭:3 - 4m)の土壌環境とアーバスキュラー菌根菌の解析を行なった。土壌水分含量は,深い泥炭>中間>浅い泥炭であり,アーバスキュラー菌根菌の胞子密度は,深い泥炭<中間<浅い泥炭であった。栄養繁殖苗に対する市販のアーバスキュラー菌根菌資材の接種試験を,現地と国内で並行して実施し,追跡調査を行なっている。
(3) 栄養繁殖苗の生存率向上:マザーパームよりサッカー(吸枝)を切除した際の切除面の清浄化,発根促進処理の効果に関する現地試験に着手した。先に実施した現地試験の解析からは,小型のサッカーを水路に浮かべた筏状苗床で4週間程度育した場合の圃場での生存率が高いと考えられた。また,水耕栽培の実験からは,サゴヤシ幼植物は貧栄養条件でもカリウムの欠乏が生じ難いと考えら得る結果が得られている。

Reason

上述のように,現地の協力大学ならびに企業の研究施設,研究者,生産施設と技術者との連携により,現地調査,試験実施,採取した試料の国内外での解析も順調に進んでいる。2024年9月マレーシア・クチンで開催された第15回国際サゴシンポジウムを本課題代表者が会長を務めるサゴヤシ学会が共催,所属機関である名古屋大学が後援し,基調講演1件,6件の口頭発表をもって成果発表による貢献が可能となった。また,代表者がFAO事務局,ASEAN事務局,JICA本部との窓口となって国際研究と集会の連携を強化できた。それらの成果を背景に,協力大学の助教授を,代表者がセンター長を務める部局の自助努力として2025年2 - 3月に招聘し,次年度以降の研究推進に向けた準備を加速してきた。
このような進行状況から,おおむね順調に進呈していると判断する。

Strategy for Future Research Activity

次の3項目,(1) 難発芽種子の発芽勢改善等による移植苗育成技術の確立,(2) 移植苗への有用微生物等の処理による生育促進,(3) 栄養繁殖苗の生存率向上,のそれぞれの下に小項目を以下のように設けており,現地の環境を考慮しながら推進する。
1-1) 稔性種子生産に向けたポリネータ動向調査,有核果実からの種子を用いた実生生産;1-2) 人工授粉技術採用(ポリネータ定花性を考慮して異なる樹々の両性花に受粉,受粉相性を調査して推進);1-3) 種子発芽率を高めるプライミング方法の検討
2-1)実生苗に市販の有用微生物資材を接種した材料の追跡調査,実生の形態的変化,光合成能を評価;2-2)土着の有用微生物種の群集解析,選抜,培養(次世代シーケンサーを用いたアンプリコン解析で微生物叢を解明して推進);2-3)選抜した有用微生物群を実生苗に接種,育苗期間中の枯死率,形態的変化,光合成能,各部位の養分含量を分析・評価(菌体懸濁液を培土に灌注する方法とサゴヤシ根を懸濁液に浸漬する方法におよる効果を比較);2- 4)有用微生物接種苗の泥炭層の深さが異なる土壌での成長把握
3-1)サゴヤシに内生する有用微生物の選抜,培養;3-2)サゴヤシ樹皮灰を切除面に塗布する物理処理(木灰利用による切除面腐敗防止を検討推進)
現地プランテーション圃場,協力教育研究機関ならびに協力企業の圃場や研究施設の活用,また,自助努力による研究者の受け入れと連絡試験実施の強化も交えて各課題に取り組む。
