Outline of Annual Research Achievements

動物ミトコンドリアでは通常の普遍暗号とは異なる変則暗号を用いていることが知られている。棘皮動物であるウニのミトコンドリアでは、通常LysをコードするAAAコドンがAsnに、通常ストップコドンであるUGAがTrpに、通常ArgのコドンであるAGR(R=A, G)がSerにそれぞれ変化している。これらの変則暗号の成立基盤を解明するために、ミトコンドリアtRNAを単離精製し、質量分析法を用いてアンチコドンの修飾を調べた。キタムラサキウニの卵巣より抽出したtotal RNAを陰イオン交換カラムで粗精製した後に、往復循環クロマトグラフィー法で7種類のミトコンドリアtRNAを精製した。得られたtRNAを各種RNaseで消化した後、キャピラリー液体クロマトグラフィー質量分析法で解析を行った。tRNA^<Asn>はアンチコドン2字目にpseudouridine(Ψ)が見つかり、この修飾がAAAコドンをAsnに解読するしくみであると考えられた。tRNA^<Ser>のアンチコドン1字目から7-methylguanosine(m^7G)が見出され、この修飾がAGRコドンをSerに解読することを可能にすると考えられる。tRNA^<Trp>のアンチコドン1字目から5-taurinomethyl-2-thiouridine(τm^5s^2U)が見つかり、UGAコドンがTrpに解読する機構が明らかとなった。さらに、tRNA^<Lys>から新規の修飾体が見出されたので、現在、その化学構造の同定を行っている。
