Outline of Final Research Achievements |
In the present study, we analyzed an enhancer sequence composed of Spodoptera frugiperda nucleopolyhedrovirus polyhedorin 5’-UTR for improvement of Bombyx mori cell-free protein synthesis system. As a result, we found important sequence of 18 base length composed 3'-terminal region. A protein synthesis activity using the sequence as enhancer was 1.3-fold greater than that of using usual enhancer. We constructed cell-free translation and analysis system with co-expression of activation protein using organic solvent resistant tyrosinase as a model protein. Furthermore, we also constructed membrane-binding protein expression and analysis system using thermostable laccase as a model protein.
