Outline of Annual Research Achievements

昨年度までに実施した旧来法で培養困難なルーメン細菌の分離・培養で得られたルーメン細菌129菌株から9菌株を選択し、農業分野の温室効果ガスに関するグローバル・リサーチ・アライアンス(GRA)の家畜分野が実施する国際共同プロジェクト(Hungate1000)においてゲノム解析を実施した。その結果、7菌株(Anaerovibrio sp. RM50、Lachnospiraceae bacterium G11、Lachnospiraceae bacterium G41、Lachnospiraceae bacterium RM5、Prevotella sp. RM4、Ruminobacter sp. RM87、Ruminococcaceae bacterium P7)の全ゲノム配列が決定され、公開された(http://genome.jgi.doe.gov/TheHunmicrobiome/TheHunmicrobiome.info.html)。これらの菌株のゲノム配列には、炭水化物(デンプン、セルロース、キシラン等)の加水分解酵素をコードする遺伝子が含まれていた。また、GRAの別の国際プロジェクト(Global Rumen Census)の研究成果で示された主要ルーメン細菌トップ50に、これらの菌株のうち、Ruminobacter sp. RM87とRuminococcaceae bacterium P7が含まれていたことから(1)、牛などの反すう家畜の消化生理で重要なルーメン細菌の主要メンバーとなっている2菌株の分離と全ゲノム配列の決定が達成されたことが明らかになった。
