Outline of Annual Research Achievements

難培養性菌種を含む歯肉縁下プラーク細菌は、歯周炎の原因となる細菌群であり、それらは、歯周炎の病態のみならず全身疾患の病態に影響すると予想し、培養困難な菌種を純粋培養を用いずに増殖させ、血清学的な検出を行えるように抗体を得ることを計画した。
難培養性菌種を含む歯肉縁下プラーク試料の提供者の歯周炎病態の臨床的観察によれば、急性症状を伴わず、緩徐に病状が進行する病態であることが確認された。
マウス腸管への難培養性細菌の定着を検出するための群集解析に用いるオリゴヌクレオチドプライマーの塩基配列については、Human Microbiome Projectで用いられているV3およびV5プライマーのそれを採用することとした。バイオインフォマティクスの手法を用いて、Human Microbiome Projectのデータベース中の塩基配列から、健康な歯肉縁下プラークには存在しないが、慢性歯周炎患者の歯肉縁下プラークに存在する細菌群集を検出することができた。また、国外研究者によって慢性関節リウマチとの関連性が指摘されたPrevotella copriの腸内定着に関連して、同菌の追加検索を行った。慢性関節リウマチに罹患していない提供者に関しては、同菌の口腔内定着は調べた限りにおいて検出することはできなかった。
マウス腸管における口腔細菌の定着や増殖、抗体の作製については、異動に伴い実施困難となったため、実施できなかった。
