Outline of Final Research Achievements |
We have carried out developments for technological tools to investigate single-molecule gene expression dynamics in single cells of higher organisms in a high-throughput manner. To realize this, we have developed a light-sheet illumination fluorescence microscopy that enables 3D volumetric imaging within the whole volume of thick cells set on a multi-well plate. In addition, we have developed a probing technique to detect mRNA and protein expressions at single molecule levels. Through analysis of gene expression dynamics in Saccharomyces cerevisiae cells for multiple gene targets, we have found a common feature in mechanisms on gene expression noise.
