The transcriptional coactivator PGC-1alpha is implicated in the regulation of body mass and insulin sensitivity; however, its contribution to these conditions is unclear. Two recently identified PGC-1alpha variants are transcribed from an alternative promoter and induced by exercise in skeletal muscle. Here we show that mice lacking these variants develop age-dependent obesity and insulin resistance. Although the total PGC-1alpha abundance in skeletal muscle was unaltered under static conditions, exercise-induced increases in total PGC-1alpha and energy expenditure were attenuated in the mutant mice. These results suggest that PGC-1alpha variants are responsible for exercise-induced PGC-1alpha upregulation, which in turn plays an important role in controlling fat mass and insulin sensitivity via the regulation of energy expenditure during exercise.
