Outline of Research at the Start

木質細胞壁を構成するリグニンの分子構造は、植物種・組織・細胞・細胞壁層レベルで複雑に変化する。このようなリグニンの構造多様性は、植物の進化や細胞壁の機能分化とよく相関した細胞壁の本質的特徴であると同時に、脱炭素社会構築を担う木質バイオマス利用の重要阻害要因でもある。本研究では、リグニンの化学構造を様々に改変した組換え植物の解析に基づき、細胞壁の機能及び物性、そしてその基盤となる細胞壁の固体高次構造に及ぼすリグニンの寄与の解明に挑む。

Outline of Annual Research Achievements

木質細胞壁を構成するリグニンの分子構造は、植物種・組織・細胞・細胞壁層レベルで複雑に変化する。このようなリグニンの構造多様性は、植物の進化や細胞壁の機能分化とよく相関した細胞壁の本質的特徴であると同時に、脱炭素社会構築を担う木質バイオマス利用の重要阻害要因でもある。本研究では、リグニンの化学構造を様々に改変した組換え植物の解析に基づき、細胞壁の機能及び物性、そしてその基盤となる細胞壁の固体高次構造に及ぼすリグニンの寄与の解明に挑む。リグニンのサブストラクチャー組成を網羅的かつ体系的に変化させた組換えモデル植物の細胞壁試料を作出し、化学分析・高磁場核磁気共鳴、X線、電子顕微鏡観察等により、細胞壁の一次構造と固体高次構造の関連付けを行う。2024年度は、イネのリグニン生合成に寄与する遺伝子群の解析を行い、それらを標的としたゲノム編集により、リグニンのサブストラクチャー組成を改変した新たなイネ変異株の作出に成功した。また、これまでに作出したリグニンの芳香核組成及びリグニン-多糖間の架橋に寄与する細胞壁結合型フェルラ酸量を改変したイネ株について、固体NMR、X線回折及びX線散乱を用いた固体高次構造解析を行い、特に細胞壁結合型フェルラ酸量がイネ細胞壁の固体高次構造に顕著な影響を与えることを明らかにした。さらに、種々の植物種におけるリグニン生合成機構の解明、代謝工学、生物分解等を目的とする共同研究において、リグニン構造解析やイメージング解析の実験も行った。これらの成果の一部を纏め、論文発表及び学会発表を行った。

Reason

当初の計画以上に、多数のリグニン及び細胞壁結合型フェルラ酸改変イネ株(C4H、C3'H、CAld5H、HCALDH、PMT、LAC及びそれらの多重ノックアウト株)の作出に成功し、過去に作出したリグニン改変組換えイネとともに、細胞壁の化学構造及び超分子構造の解析を進めることができた。また、種々の植物種におけるリグニン生合成機構の解明、代謝工学、生物分解等を目的とする共同研究において、リグニン構造解析やイメージング解析の実験で貢献した。これらの成果の一部を纏め、9件の論文発表と17件の学会発表を行った。以上のことから、本課題は、当初の計画以上に進展していると考えている。

Strategy for Future Research Activity

2025年度は、これまでに作出を完了しているイネおよびシロイヌナズナ株について、細胞壁の固体高次構造データの更なる収集と一次構造との関連付けを進めるとともに、現在作出を進めているリグニン修飾構造改変イネ株やそれらの複合多重変異株、ラッカーゼ変異株の解析に展開する予定である。
