Outline of Research at the Start	本申請の最終目的は、ガラス化した未成熟卵および受精卵を用いてゲノム編集胚を作成し、これらガラス化卵由来のゲノム編集豚を世界で初めて作成することである。ガラス化による卵子のDNA損傷が胚発生に悪影響を及ぼすため、まず、ガラス化処理が起因する DNA 損傷を軽減する方法を検討し、胚発生能を改善するための新規手法を開発する。次いで、至適化されたガラス化保存法を利用して、ゲノム編集豚の生産に取り組む。ゲノム編集のターゲット遺伝子として成長ホルモン受容体を選定し、受精卵エレクトロポレーション法により当該遺伝子の変異誘導を試み、最終的に小型化豚の生産によりゲノム編集の成否を判断する。
