Outline of Research at the Start

転写因子SOX10は、悪性黒色腫特異的がん遺伝子MITFの発現を調節する。MITFががん遺伝子であるため、悪性黒色腫組織内には「SOX10/MITF高発現の増殖能が高い細胞」が多く存在する。一方、近年のsingle cell RNA-seqの解析から「SOX10/MITF低発現の浸潤能が高い細胞」、「SOX10高発現/MITF低発現の細胞」も存在することが明らかになってきた。しかし、「SOX10/MITF低発現細胞」については依然として未解明な部分が多い。本研究では、SOX10の機能解析により、がん転移・薬剤耐性などの患者予後の悪化に繋がる「SOX10/MITF低発現細胞の特性」を明らかにし、「悪性黒色腫の不均一性と腫瘍悪性化進展」を理解することを目指す。
