Outline of Research at the Start

本研究の目的は、基底前駆細胞に着目することで、喘息の多様な病態を解明し、治療に結びつけることである。そこで、まず気管支喘息患者における基底前駆細胞の役割を組織検体で解析する。低侵襲に組織検体を得る方法として、外科の手術検体の余剰検体を用いて免疫染色を行い、基底前駆細胞の発現と患者背景を比較する。次にヒト検体より基底前駆細胞を既報の方法で選択培養して、シングルセルRNA-seq解析に用いるとともに、上皮細胞に分化していく様子を観察する。さらに分化誘導因子の産生細胞を特定した後、慢性気管支喘息マウスモデルを作成し、治療実験を行う。
