Outline of Research at the Start

本研究では、EBV感染細胞を特異的に障害する薬剤の開発を目指す。BARTプロモーターの下流にルシフェラーゼ遺伝子を挿入したベクターを上皮細胞に導入し、薬剤ライブラリーから阻害剤を探索する。同定した化合物において、BART miRNAs発現に対する影響と抗ウイルス効果を、EBV感染細胞と非感染細胞で確認する。次に、免疫不全マウスで抗腫瘍効果を確認する。候補薬剤が、どの転写因子がBARTプロモーターへ結合することを阻害しているかを、変異プロモーターを作製することで明らかにし、転写因子と阻害剤の直接作用を詳細にし、抗EBV薬を開発する。
