Outline of Research at the Start

ヒトES細胞は、軟骨分化が可能である。再生軟骨による気道再建は、自己細胞を継代培養し再生軟骨が得られている。この手法は、自己組織への侵襲があり、初代培養の高額な費用と長期の培養期間が必要となる。本研究の目的は、ヒトES細胞軟骨由来の組織工学的再生軟骨と細胞シート工学によるハイブリッド再生軟骨を用いた、異種移植による気道再建を実現することである。そのために、①ヒトES軟骨から軟骨細胞を単離・継代培養し軟骨細胞シートを作成、②家兎の進化型生分解性足場材料の開発、③軟骨細胞を生分解性足場に播種し軟骨細胞シートで被覆し軟骨再生、④家兎でヒトES細胞由来軟
骨による異種気道再建を実証する。
