Outline of Research at the Start

我々は消化管癌外科治療において、狭窄、瘻孔形成予防に対して粘膜上皮や筋層の再生医療的治療法を開発し、臨床応用まで行った。上皮層と筋層に対する一定の効果を得た一方で、線維化や難治性瘻孔に対して真の消化管再生には自己修復能を持つ上皮層と筋層の融合が必須である。本申請では、臍帯由来上皮幹細胞と間葉系幹細胞に着目し，分化誘導筋芽細胞と分化誘導上皮細胞を細胞培養で増殖させ、三次元的な消化管構造を生成する、新規の細胞培養手法を開発する。さらに、この組織構造を実験的モデル動物に移植して組織の形態学的および機能的な変化を定量化することで、再生組織の適合性と持続性の新たな評価系を確立する。
