Outline of Research at the Start

ヒトiPS細胞から中枢神経細胞を培養下で分化誘導し、培養液に種々の麻酔薬を添加して麻酔薬の神経細胞への影響、特に神経毒性と神経保護活性の詳細を明らかにする。形態面の解析として、Trypan Blue染色、TUNEL染色を行う。同時に、機能面の解析としてFluo-4を用いたCa2+イメージング法を用いた解析を行い、形態的に生存していても脱落してしまう生理機能を順次検証していく。将来的には、ミトコンドリア病患者等の遺伝性疾患のiPS細胞で同様の実験を行い、それら患者の脳保護に最適な麻酔薬選択の一助とする。
