Outline of Research at the Start

二分脊椎症は先天性の異常である。特に、脊髄髄膜瘤は指定難病118番に指定され、生涯にわたる医療ケアを要する。申請者は、患者の全ゲノム解析により、遺伝子変異を同定した。そして、それら変異遺伝子がコードする転写因子の活性に異常を認めた。さらに、遺伝子変異が脊椎の形成不全をきたすことを明らかにした。以上の成果を踏まえて、本研究は申請者が同定した転写因子に着目し、二分脊椎症の責任遺伝子を同定する。そのために、変異モデル動物における遺伝子発現パスウェイを解析する。本研究成果は、責任遺伝子を標的とした二分脊椎症の予防法や根治療法の開発に波及する。
