Outline of Research at the Start

HPV陽性の中咽頭癌は、HPV陰性の癌より放射線や化学療法への感受性が高い事が知られているが、その機序は不明である。培養細胞にHPV16 E6を発現させるとPARPが強く発現し、著しいポリADPリボシル化が誘導される。PARPはDNA修復酵素の一つであるが、過剰なポリADPリボシル化は、PARP活性の低下とアポトーシスを誘導する。従ってHPV陽性の中咽頭癌では、E6の発現が過剰なポリADPリボシル化を誘導し、PARP活性を低下させ、抗癌剤や放射線への感受性を亢進させていると考えた。本研究は、HPV感染によるPARP活性の低下が治療感受性に与える影響を明らかにし、さらに臨床応用への発展を目指す。
