The transcriptional activation of the zygote after fertilization is a crucial stage of preimplantation development in mammals. Recent studies have identified pioneer factors directly involved in the activation of stage-specific genes and endogenous retroviruses including ERVL. However, how these pioneer factors themselves are activated remains unknown. The aim of this research is to identify and characterize the ‘initiator’ proteins that activate these pioneer factors to further our understanding of the transcriptional activation in early embryos.
