本研究の目的は、HMG-CoA還元酵素阻害薬の抗白血病効果の機序を解明することである。
その独創性は、スタチン投与下でのAML細胞株のCRISPR knockout screeningを行い、その結果に基づいて解析を行う点にある。スクリーニングは実施済みであり、statinによる細胞死に関連すると考えられる分子を特定し、また、pathway解析により、関連する経路を特定した。
本研究は、スタチンが白血病細胞死をきたす分子機序を、上記で特定した遺伝子群やpathwayの観点から解明する試みである。既存化学療法の効果を高めるスタチンの投与法の開発のみならず、新規の分子標的薬開発につながる可能性がある。
