Macrocyclic peptides have received considerable interest in drug discovery as a promising drug modality, but their low cell permeability has restricted their use to extracellular targets. Given that many drug targets reside inside cells, a generally applicable strategy for the discovery of cell-permeable and bioactive peptides is a strong demand. This research aims to address this challenging task by developing a novel mRNA display-based system, employing a pseudo-natural product library that possesses strategically positioned backbone N-methyl groups.
