胆道癌は本邦において5年生存率は膵癌に次いで低く、予後不良な癌腫であり、発癌のメカニズムや有効な治療法も少ない。ARID1Aは胆道癌において高頻度に遺伝子変異がみられ、、また癌抑制遺伝子としての機能が示唆されていることから、我々はARID1Aが胆道癌の形成において腫瘍抑制的に働いているのではないかと考えた。またARID1Aの変異が、多くの癌腫でPI3K-Akt経路の変異と重複し、ヒト胆道癌における遺伝子解析においても、PI3K-Akt経路との同時変異が多く、関連性が示唆される点に着目し、ARID1Aの胆道癌形成における役割を遺伝子改変モデルマウスを用いて明らかにすることとした。
