| Outline of Research at the Start |
全酵素中の1/2を占めると言われている金属酵素は様々な疾病に関連しているものの、臨床で使用されている阻害剤の数は限られています。そこで本研究では、金属酵素に対する新しい不可逆阻害剤を開発することを目的とします。具体的には、金属配位部位を有する求電子フラグメントを用いてスクリーニングを行い、金属酵素の活性を低下させる化合物を探索します。本系の実証として、活性中心に鉄(II)イオンおよび求核性アミノ酸残基(Lys等)を有するKDM4B(リジン脱メチル化酵素)に対するスクリーニングを行い、不可逆阻害剤の開発を行います。
