シスプラチンなどの白金製剤は「古い」抗がん剤であるが、現在も多くの標準治療で使われ、臨床的に重要な抗がん剤である。一方、臨床における課題として、腎障害など重篤な副作用があげられる。研究代表者はこの強い副作用が、タンパク結合率が高いとされる白金製剤が、実は血中に投与された直後にはタンパク質と結合せず、多量に存在する遊離型が一過性に全身に分布することで生じている可能性を見出している。本研究では、白金製剤のタンパク結合割合と副作用/薬理作用との関係を、新しい分離定量システムを用いて改めて評価し、白金製剤の毒性と効果発現のバランスを制御し得る“新しいがん治療アプローチ”を提唱する。
