Outline of Annual Research Achievements

減数分裂期ではDNA複製に伴って姉妹染色分体が接着(コヒージョン)した後、二回の連続した染色体分配が起きる。第一分裂では姉妹染色分体は同一極へと分配され、続く第二分裂は両極へと分配される。第一分裂期では、Sgo1によってセントロメア領域の接着が保護され、この接着は続く第二分裂を正確に行うために必要である。Sgo1は動原体因子Bub1に依存してセントロメアへ集積して機能することが知られている。また減数第一分裂期特異的な動原体因子Moa1(Meikin)とその下流因子Plo1キナーゼにも、セントロメアの接着を保護する機能があることが知られていたが、その詳細な分子メカニズムはこれまで不明であった。本研究により、減数分裂期においてMoa1-Plo1がセントロメア領域の動原体因子Spc7をリン酸化し、その結果Bub1を動原体へ集積し、Sgo1の十分な局在化に寄与することが示された(Genes to Cells 2017)。
正確な染色体分配を保証するためには、複製された姉妹染色分体が分裂中期まで接着され、細胞の両極から伸長するスピンドル微小管により正しく捉えられる必要がある。姉妹染色分体間の接着は、リング状のタンパク質複合体であるコヒーシンによって担われている。一方、ヒストン・キナーゼHrkl/Haspinは、セントロメアへ局在し、姉妹染色体の両方向性結合を確立させる働きがあるAuroraBキナーゼをセントロメアへと呼び込む働きがある。本研究では、コヒーシンサブユニットのひとつであるPds5が、コヒーシン制御因子およびHrkl/Haspinと直接相互作用することを示した。すなわち、Pds5は保存された結合モチーフを使って、細胞内の複数の制御因子と相互作用することで、正確な染色体分配を保証していることを明らかにした(Curr Bio1 2017)。
マウスの生殖細胞特異的な発現をする因子のスクリーニングで得られた染色体軸因子Iho1が、染色体軸で組み換えに関わっていることを明らかにした(Nature Cel1 Bio1 2016)。

Reason

減数分裂の動原体の制御因子Moal(Meikin)によるセントロメアの接着保護機構の一端が明らかになった。染色体の二方向制御因子Haspinのセントロメア局在機構が明らかになった。マウスの生殖細胞での染色体組み換え制御機構の一端が明らかになった。

Strategy for Future Research Activity

今回の分裂酵母における解析から、Moa1-Plo1依存的な接着保護機構には、Sgo1の局在制御以外に別の経路があることも分かってきた。これに関しては、Moa1-Plo1がコヒーシン複合体のサブユニットのひとつであるRec8をリン酸化することで、Sgo1依存的にコヒーシンが保護されやすくなるという予備的な結果を得ている。今後は、その分子機構を詳細に解析する。また、マウスにおいても同様の制御があるか調べる。
マウスの生殖細胞で、相同染色体のペアリングに関わる因子の同定を進める。特に、我々が今までにRad21Lコヒーシンがペアリングに関わることを明らかにしてきた。今後は、Rad21Lと複合体を形成する因子を探索し、ペアリングの分子機構の解明を目指す。
