Outline of Annual Research Achievements

本研究では、自己免疫疾患の一つ関節リウマチ（以下、RA）と関係する蛋白質の糖鎖修飾を効率よく同定する手段として、複数の糖蛋白質についての同時糖修飾探索系の確立を目的とした。これまでに健常者の末梢血単核球(peripheral blood mononuclear cell(PBMC))を蛋白質源として、アスパラギンに付加されるN型糖鎖を認識するレクチンを用いて、N型糖鎖の存在を確認したことにより、細胞質蛋白質にも糖蛋白質が存在することを明らかにした。この結果により、レクチンをリガンドとしたアフィニティーカラムにより、特異的な糖鎖構造を有する糖蛋白質を精製可能であると考えられた。26年度にPBMCの細胞質蛋白質中の糖蛋白質の精製を予定していたが、研究代表者が病気により、長期休職をする状況となり、研究の継続が難しくなったため、糖蛋白質の精製には至っていない。
