Outline of Final Research Achievements |
Poly(ADP-ribosyl)ation is a post-translational modification of protein in which the ADP-ribose moiety of NAD+ is transferred to amino acids in proteins, altering their structure and function. This protein modification is catalyzed mainly by a nuclear poly(ADP-ribose) polymerase (PARP) in response to several stress, most notably DNA damage. PARP orthologs are found in mammals, plants, metazoans, protists and filamentous fungi, but not in yeast, while PARG homologs are identified in all eukaryotes, besides fungi. Here, we searched for a parg orthlog in the genome sequence of a model fungus A. nidulans. First, we picked up seven genes as candidates of a fungal parg gene. Proteins coded by the genes were successfully expressed as soluble proteins using Escherichia coli expression system. Among them, only one protein was found to hydrolyze poly(ADP-ribose). This is the first report for identification of PARG in filamentous fungi.
