Outline of Research at the Start

刺激を加えたSVFからEVを精製して内部に含まれるmicroRNAを解析すると同時に、軟骨細胞への作用を調べる。SVFに含まれる各細胞群をFACS解析で明らかにし、軟骨細胞を炎症性サイトカインであるIL-1βで刺激する実験系にEVを添加した際のMMP13, ADAMTS4,5,9mRNA発現への影響を定量PCRおよびアグリカン分解を検出するウエスタンブロッティングにて検討する。
さらに細胞実験で効果が得られたEVについては雄性SDラットまたはICRマウス実験的OAモデル膝関節投与実験をおこない、OA抑制効果が得られるかについて、Safranin-O染色およびアグリカン免疫染色にて検討する。
