Outline of Research at the Start

アンモニアは生体に有害であるが、アミノ酸代謝などで必然的に生成される化学物質である。本来生体はアンモニアの解毒機構を備えているが、先天的要因などでこの解毒機構が働かない場合、血中のアンモニア濃度が異常に高くなり高アンモニア血症となる。現在いくつかの治療薬が存在するが緊急時に十分な対応はできない。そこで本研究では、PIKfyveというリン酸化酵素を阻害することでアンモニアを細胞内の特定の構造体（リソソーム）に隔離させ高アンモニア血症を治療できないか検討する。PIKfyve阻害薬は、ALSの治療薬としての可能性もあり盛んに研究されている。本研究はその薬物作用に対する基礎的知見も提供する。
