Outline of Research at the Start

本研究は、MSI（マイクロサテライト不安定性）陽性胃癌の分子機構を解明し、新たな治療戦略を構築することを目的とする。MSI陽性腫瘍はMLH1の異常高メチル化、CIMP-high、野生型TP53の保持といった特異な形質を示すが、それらの獲得機序や相互関係は未解明である。申請者は、MLH1の不活化に関わる転写制御の破綻や、野生型p53が機能を失わずに発癌に至る仕組みに注目してきた。本研究では、①p53の機能抑制機構、②MLH1不活化の分子機構、③p53の活性を引き出す新たな治療戦略の検証を通じて、MSI腫瘍の本態とその脆弱性を明らかにすることを目指す。
