Outline of Research at the Start

造血器腫瘍に対して著効を発揮したキメラ抗原受容体導入Ｔ細胞輸注療法の固形がんに対する適応拡大が期待されている。しかし、固形がんにおいては、輸注CAR-Tの低浸潤性やがん微小環境に於ける免疫抑制機構（Treg、免疫抑制分子、免疫抑制性代謝産物）がその有効性を損なっている。加えて、がん微小環境の貧栄養・低酸素状態における慢性的・持続的な抗原刺激によりCAR-Tの機能障害と疲弊をもたらすことも明らかになってきた。したがって、この様な状況下でも優れた抗腫瘍活性が発揮・維持できるCAR-Tを作製することが固形がんに有効なCAR-T輸注療法においては必要である。
