Outline of Research at the Start

アルツハイマー病の危険因子であるApoE4は人種を超えて確立されているが、その機序は不明である。本研究ではApoE4がタウ蛋白のシード依存性凝集にどのように影響するかを解析するため、ApoE2,E3およびE4ノックインマウスにアルツハイマー病脳をシードとして脳注入しタウ伝播凝集状態の解析後、シングルセル解析およびRNAsequencingを行いApoEアイソフォームがタウ凝集へおよぼす分子機構を明らかにする。更にタウ注入ApoE4モデルマウスに保護的であるとされるApoE2をウイルスベクターに経静脈投与し運搬させ遺伝子治療をめざす。
