Outline of Research at the Start

近年、NSCLCの新規遺伝子異常としてPTPN11変異が3%認められること、KRASとの共変異が30%であることが分かった。SHP2阻害剤は変異SHP2に阻害効果がなく、PTPN11変異陽性疾患では別の治療戦略が必要である。本研究では、ゲノム編集によりPTPN11変異の有無のみ異なるNSCLC細胞株を樹立し、変異SHP2によるメチル化調節機構の有無を解明する。そして、PTPN11変異陽性NSCLCに対する治療戦略の改良につなげる。
