Outline of Research at the Start

本研究では、申請者らが独自に作製したDMKN欠損マウスの胎仔期にDMKNリコンビナント蛋白投与やDMKN発現自己末梢血幹細胞により、新生仔期にDMKN強制発現骨髄細胞を移入する。これにより、出生後に魚鱗癬様皮膚と皮膚バリア機能の改善が得られるか、その効果が持続するかどうかを検証する。胎仔へのDMKN投与は、３種類の異なる投与法（循環系内、胎仔腹腔内、胎仔皮下）で施行する。皮膚のDMKN発現量と皮膚症状の改善度から、標的分子の高い導入効率と免疫寛容が得られる投与法を確立する。
