Outline of Research at the Start

本研究では、肝芽腫におけるがん幹細胞の性状を明らかにし、治療標的となる分子群の同定を目的に、摘出組織や既知の肝芽腫 cell line から、がん幹細胞の長期培養法を検討し、特に幹細胞性 sphere 形成能と分化誘導能を有する細胞群の単離・樹立を介して、分化誘導モデルの作製を行う。このモデルを用いて、分化誘導刺激で変化する細胞内活性化分子ネットワークを融合プロテオミクスによる分子プロファイリングで詳細に解析し、幹細胞維持と分化誘導スイッチングにおける分子メカニズムの一端を明らかにする。得られた情報をもとに、肝芽腫がん幹細胞をターゲットとした分子標的治療戦略に有用となるデータ収集を目標とする。
